What size? Depends on what resolution you want the game at... (same with the characters) backgrounds... anything really

If you want my opinion, the best way to figure it all out... is just do it

I experimented with AGS for over a month, trying different things graphics and so on and so forth... THEN I started on my game...

practice makes perfect, and there is no substitute for experience.

I mean the size of the Pictures you are Creating for example in paint.
Are you really creating a picture in 320 x 240 ???? or are you creating a picture width a size of for example 640 x 480? Just for resizing it after you finished it? The Character for example indiana jones is so small... How do you paint him??? I ask for the details... Shadows etc.?

Some people can actually draw characters that small. But usually if you're making a 320x200 game, you draw the background in that resolution. Maybe one or two people here resize from a larger image, but I doubt it.
