This is very, very annoying.

So I start my trusty PC up yesterday and lo and behold I get some message about it not being able to load my profile and "DETAIL - Error performing inpage operation."
It loads Windows ok, except it gives me some basic default profile. All my emails, bookmarks, desktop icons and files, startup apps etc... are gone. Everything is reset.
Let me rephrase that. They aren't actually gone. The folder containing my profile and all associated files is still there, it's just not actually loading them.
Not only that, but even if I set everything up again it resets when I reboot.

Help me. Please. Help me.

did you try a system restore to a time before this started happening?  have you istalled anything new latley?  maybe try settting up a new profile under a different name but with the settings the same?

Sound pretty hinky to me.  Kind of like that time I came ho,e from work, turmed on my computer and discovered that windows was simply gone (thus causiing me to upgradde to Win XP).  Other then those suggestiions, I'm not sure what it could be, but I hope you getted sorted  :-[
