I can't prepare the spells in the dungeon, in the house, outside, or at the bottom of the mountain, and if I cast them in the dungeon, they act on me and I die.

Every time I cast a spell in the dungeon, it works on me and I die. So that's not the way to do it.

I've found out how to do this. To answer my own question, in case it helps anyone else:

Once you have all the ingredients for a spell, you need only make sure they are in your inventory (and not under the bed) and then click on the spell in the book with the "CAST" cursor. You don't actually need to carry out the actions described in the spell.

When can I prepare the spells?

I have everything I need except the stone. I have the wand. When I go down into the dungeon and try to cut the cactus, it says "No"; if I use the cactus on the knife, it says "You can't do that yet."

Do I have to collect all of the ingredients before I can prepare the spells, or do I have to wait for Manannan to be doing something in particular? At the moment, he is sleeping.

It's a while since I played it, but there is a club you go to and the doorman is standing outside. I think you have to ask the taxi driver to take you there. If you don't have the option of going there yet, you need to find a clue that suggests you should go there.

Either I get killed if I walk into the room where he is because I've discovered the "magics", or I get killed when I am nearly at the bottom of the mountain because I'm "not allowed out here".

If I remember rightly, I had the same problem. This game seems to have been released without a winsetup file. Winsetup files might not have been around when the first in the series was made, but that's no excuse for not providing one for a recent release.

stop Mannanan from killing you every five seconds? I've picked up a whole load of things from the house, washed the floor, emptied the chamberpot and left, but as soon as I start to walk down the mountain, the wizard appears and kills me.

To give an object to a person, right-click on the person, then select the object from your inventory. The same applies to using an inventory object on something in a room, but, obviously, only works where there are hotspots.
